Emergency services were called to the scene at around 6.30am, and the A303 was closed in both directions between Longbarrow roundabout and Deptford junctions.
Police say a woman was "seriously injured".
Fire crews from Salisbury, Warminster and Basingstoke attended the scene, as well as Wiltshire Police and the ambulance service.
An earlier statement from Wiltshire Police said: "We have closed the A303 in both directions between Longbarrow roundabout and Deptford junctions following a serious collision involving a lorry and a van.
"The closure is likely to be in place for some time while we deal with the scene and ambulance colleagues deal with a woman who is seriously injured.
"Please avoid the area and seek an alternative route for the duration of the morning.
"Thank you for your patience."
